Resumo O estudo da diversidade genética de Theobroma speciosum é importante, pois pode ser utilizado em programas de melhoramento, uma vez que a espécie está intimamente relacionada a espécies de grande valor econômico, como Theobroma cacao (cacau) e Theobroma grandiflorum (cupuaçu). Assim, o objetivo deste trabalho é caracterizar a diversidade genética intra e interpopulacional de Theobroma speciosum em populações naturais na Amazônia brasileira. Foram selecionados e genotipados 90 indivíduos de T. speciosum provenientes de quatro populações situadas em diferentes estados da Amazônia Legal. Os dados foram obtidos por meio da análise de microssatélite e a diversidade genética foi caracterizada através do número de alelos, número de alelos privados, índice de fixação e heterozigosidade observada e esperada. A análise bayesiana, a AMOVA e PCOa foram utilizadas para revelar a estrutura genética molecular das populações, através dos programas Structure e GenAIEx 6.5, respectivamente. Todas as populações estudadas apresentaram níveis de diversidade gênica, contudo, as populações AUR, API e MAC apresentaram grande similaridade, enquanto a população de Rio Branco apresentou maior heterozigosidade e menor endogamia do que as outras, se tornando um ponto de refúgio na Amazônia, e a mais importante população para a conservação de T. speciosum.

Abstract The genetic diversity of Theobroma speciosum is important because its use in breeding programs, once the species is closely related to species of great economic value such as Theobroma cacao (cocoa) and Theobroma grandiflorum (cupuaçu). Thus, the objective of this work is to characterize the intra and interpopulational genetic diversity of Theobroma speciosum in natural populations in the Brazilian Amazon. Ninety individuals of T. speciosum from four populations localized in different states of legal Amazon were selected and genotyped. The data were obtained by fluorescence microsatellite analysis and the number of alleles, number of private alleles, fixation index, observed and expected heterozygosity were analyzed. Bayesian analysis, AMOVA and PCOa were used to reveal the molecular genetic structure of the populations, using the programs Structure and GenAIEx 6.5, respectively. All populations studied present great levels of gene diversity, although, there was a greater similarity among the AUR, API and MAC populations, while RBC population presented higher heterozygosity and less inbreeding than the others, becoming a possible refuge area in the Amazon, and the most important population for T. speciosum conservation.

RESUMO Muitas árvores tropicais possuem dossel alto e folhas não facilmente acessíveis. O uso de tecido de um órgão mais acessível (câmbio) para extração de DNA pode ser uma alternativa para estudos moleculares. Nós adaptamos uma metodologia viável para extrair DNA genômico de tecido cambial coletado no campo para avaliação com PCR. Testamos três condições de armazenamento (dois tampões e sílica gel) e quatro períodos após a coleta. Utilizamos protocolos descritos anteriormente e os testamos em três espécies encontradas em florestas amazônicas e outros biomas: Anadenanthera peregrina var. peregrina, Cedrela fissilis e Ceiba speciosa. Nosso protocolo foi eficaz na obtenção de DNA adequado para sequenciamento e genotipagem de microssatélites. Recomendamos o uso de sílica para armazenamento de longo prazo e o tampão com ácido ascórbico para curto prazo.

ABSTRACT Many tropical trees have high canopies and their leaves are not accessible. Thus, the use of tissue from a more accessible organ (cambium) for DNA extraction may be an alternative for molecular studies. We adapted a feasible methodology for extracting genomic DNA from cambium tissue harvested in the field for the assessment with PCR. We tested three storage conditions (two buffers and a silica gel) and four periods of time after harvest. We used previously described protocols and tested them on three species that occur in Amazonian forests and other biomes: Anadenanthera peregrina var. peregrina, Cedrela fissilis, and Ceiba speciosa. Our protocol obtained suitable PCR-grade genomic DNA for DNA sequencing and microsatellite genotyping. We recommend the use of silica for long-term storage and the buffer with ascorbic acid for short-term storage.

RESUMO A macaúba, Acrocomia aculeata (Jacq.) Lodd. ex Mart.), é uma das espécies nativas brasileiras com maior potencial para a produção de biocombustíveis. O objetivo deste trabalho foi analisar a diversidade e estruturação genética de macaúba para auxiliar na definição de estratégias de amostragem para bancos de germoplasma. Quarenta e seis marcadores microssatélites, dos quais sete marcadores polimórficos foram utilizados para avaliar 103 indivíduos de macaúba coletados em diferentes regiões do Brasil. Os marcadores polimórficos foram utilizados para gerar uma matriz de dissimilaridade pelo índice ponderado. A visualização da variabilidade genética foi realizada através de uma projeção 3D da matriz de dissimilaridade. Sessenta e sete destes indivíduos tiveram sua região ITS sequenciada e alinhada, e as mutações encontradas foram utilizadas para gerar uma rede de haplótipos. A distância genética média identificada entre os indivíduos foi de 76,2%, variando de 3,7 a 100%. Não foi encontrada estruturação da variabilidade genética. O sequenciamento da região ITS dos 67 indivíduos revelou quatro sítios polimórficos, definindo quatro haplótipos. Os resultados obtidos neste estudo sugerem que as populações de macaúba estiveram fortemente conectadas historicamente, indicando recente expansão populacional da espécie. Os resultados indicam que a amostragem da variabilidade genética de macaúba deve estar focada em coletas mais robustas em poucos locais, mas áreas como Caatinga e Chaco Úmido podem apresentar novas fontes de variabilidade genética, e devem ser melhor estudadas.

ABSTRACT The macaw palm, Acrocomia aculeata (Jacq.) Lodd. ex Mart., is a Brazilian native species with great potential for biofuel production. The aim of this work was to analyze macaw palm genetic diversity to structure and assist in the definition of sampling strategies for germplasm banks. Forty-six microsatellite markers, from which seven polymorphic markers were used to evaluate 103 macaw palm individuals collected from different Brazilian locations. The polymorphic markers were used to generate a dissimilarity matrix by weighted index. The imaging of genetic variability was realized by 3D projection of matrix dissimilarity. Sixty-seven individuals had their ITS region sequenced and aligned, and the mutations found were used to generate a haplotype network. The average genetic distance identified between individuals was 76.2%, ranging from 3.7 to 100%. Genetic variability structure was not found. ITS region sequencing of the 67 individuals revealed four polymorphic sites, defining four haplotypes. The results of this study suggest that historically, macaw populations were strongly connected, indicating a recent population expansion of the species. The results indicate that macaw genetic variety sampling should focus on effective collection in selected locations. Areas such as Caatinga and Humid Chaco however, could present new sources of genetic variability, and should be studied.

RESUMO: Quinze primers ISSR (entre sequências simples repetidas) foram utilizados para avaliar a diversidade genética entre e dentro de pomares comerciais de Theobroma grandiflorum (Willd. ex Spreng.) K. Schum. Para isso, foram analisados sessenta indivíduos, distribuídos nos três cultivos. Um total de 102 bandas foi amplificado, com uma porcentagem de 52,0% de polimorfismo em nível de espécie e média de 6,8 alelos por primer ISSR. A média do Índice de Conteúdo Polimórfico (PIC) foi de 0,55. Em relação aos índices de diversidade gênica de Nei (H) e de Shannon (I), os cultivos analisados apresentaram os valores: SAR H = 0,114 e I = 0,177; SSL H = 0,108 e I = 0,162 e SEC H = 0,104 e I = 0,156, considerados valores de moderados a baixos. A AMOVA revelou 34,91% da variância total entre os cultivos e 65,09% dentro deles. Os marcadores moleculares ISSR revelaram que há diversidade genética dentro de cada cultivo comercial estudado, portanto é possível selecionar genótipos superiores que poderão ser utilizados para originar cultivos mais uniformes. Esse resultado tem sido considerado de grande relevância, por fornecer ferramentas para a implementação de programas de melhoramento e delineamento de estratégias de conservação ex situ e in situ.

ABSTRACT: Fifteen ISSR (inter-simple sequence repeat) primers were used to evaluate the genetic diversity among and within commercial crops of T. grandiflorum (Willd. ex Spreng.) K. Schum. For this, 60 specimens were analyzed, distributed in three crops. A total of 102 bands were amplified, with a polymorphism percentage of 52.0% at species level and an average of 6.8 alleles per ISSR primer. The average for polymorphism information content (PIC) index was 0.55. In relation to the genetic diversity index of Nei (H), and Shannon (I), crops analyzed showed the following values: : SAR H = 0,114 e I = 0,177; SSL H = 0,108 e I = 0,162 e SEC H = 0,104 e I = 0,156, considered moderate to low values. AMOVA showed 34.91% of total variance among the crops, and 65.09% within them. The ISSR molecular markers revealed that there is genetic diversity within each commercial crops studied, thus is possible to select superior genotypes that can be used to give more uniform crops. This result has been considered of great relevance, to provide tools for breding implementation programs and design conservation strategies ex situ and in situ.

Pathogens within the Diaporthe complex cause seed decay, stem blight and stem canker on soybean, representing a serious threat for this crop species. We herein utilize worldwide sequence data retrieved from Genbank in order to assess the species boundaries between the soybean stem canker causal agents, and define whether or not they should be regarded as members of the same biological species. These studies were complemented with compatibility tests, in order to validate our findings from a biological standpoint. Species delimitation assays supported the occurrence of a speciation event between D. caulivora and D. phaseolourm var. meridionalis. A speciation hypothesis between D. aspalathi and D. phaseolourm var. meridionalis was also supported, based on three reciprocally monophyletic substitutions at locus EF1-α. Compatibility tests further validated species delimitation assays indicating that D. caulivora has developed barriers to gene exchange with D. phaseolorum var. meridionalis. Clarification of the specific boundaries of the SSC pathogens and related entities will be an important asset to future research in soybean pathology, epidemiology and breeding.

The roots of the understorey shrub Carapichea ipecacuanha (ipecac) have medicinal properties, and the uprooting of wild plants has supplied most of the world demand for this species. Although under severe population decline, C. ipecacuanha lacks legal protection. In the wild, the aerial stems of ipecac clump together to form clusters with well-defined borders. Cluster size may range from several to hundreds of aerial stems. To investigate the extent of clonality among aerial stems in ipecac clusters, we sampled 50 wild clusters (a total of 291 aerial stems) and screened them with 89 inter-simple sequence repeat (ISSR) markers. The 291 aerial stems were grouped into 42 putative clones. The clonal groups generally consisted of aerial stems from the same cluster, and there was little or no genetic differentiation among aerial stems at the cluster level. These findings suggest that strategies designed to conserve ipecac in situ should not rely upon census data, which are based on the number of aerial stems per cluster and the number of clusters per population, because such data greatly underestimate the species effective population size and genetic diversity. Our results also indicate that this species needs protection at a federal level.

β-tubulins are structural components of microtubules and the targets of benzimidazole fungicides used to control many diseases of agricultural importance. Intron polymorphisms in the intron-rich genes of these proteins have been used in phylogeographic investigations of phytopathogenic fungi. In this work, we sequenced 2764 nucleotides of the β-tubulin gene (Pp tubB) in samples of Phakopsora pachyrhizi collected from seven soybean fields in Brazil. Pp tubB contained an open reading frame of 1341 nucleotides, including nine exons and eight introns. Exon length varied from 14 to 880 nucleotides, whereas intron length varied from 76 to 102 nucleotides. The presence of only four polymorphic sites limited the usefulness of Pp tubB for phylogeographic studies in P. pachyrhizi. The gene structures of Pp tubB and orthologous β-tubulin genes of Melampsora lini and Uromyces viciae-fabae were highly conserved. The amino acid substitutions in β-tubulin proteins associated with the onset of benzimidazole resistance in model organisms, especially at His6, Glu198 and Phe200, were absent from the predicted sequence of the P. pachyrhizi β-tubulin protein.

Foram realizados estudos reprodutivos em acessos brasileiros de poaia, Psychotria ipecacuanha. Poaia apresenta heterostilia, com flores brevistilas e longistilas. O desenvolvimento do pólen foi estudado em cortes de anteras embebidas em resina. O desenvolvimento da antera seguiu o padrão normal para as dicotiledôneas, a qual apresentou quatro camadas: epiderme, endotécio, camada média, e tapete, a mais interna. O pólen apresentou-se bicelular e preenchido com amido no estágio de micrósporo. A morfologia do pólen foi estudada utilizando-se MEV. Foi observado polimorfismo polínico dentro e entre as duas formas florais. Foram identificados cinco tipos de grãos de pólen, com exina reticulada ou perfurada. Em seu hábitat natural, sabe-se que essa espécie apresenta propagação por multiplicação vegetativa, mas as características estudadas demonstraram que o mecanismo sexuado é tão importante para a reprodução dessa espécie quanto à propagação vegetativa.

The aim of this work was to carry out the reproductive studies on Brazilian accessions of ipecac, Psychotria ipecacuanha. It presented heterostyly, with brevistylous and longistylous flowers. The pollen development was observed from the sections of the anthers embedded in resin. Anther development was normal as usually observed in dicotyledones, displaying four layers: outer epidermis, endothecium, middle layer and inner tapetum. The pollen was bicellular and filled with starch at the microspore stage. Pollen morphology was studied using SEM, which showed pollen polymorphism within and between the two floral morphs. Five types of pollen with reticulate or perforate exine were identified. The characteristics showed that the sexual process was as important as the vegetative propagation for the reproduction of this species.

O objetivo deste trabalho foi estimar a diversidade genética de populações de Zabrotes subfasciatus, por meio de marcadores moleculares ISSR. Foram avaliadas 12 populações, provenientes de oito estados brasileiros, no total de 269 indivíduos. Cinco iniciadores ISSR permitiram a obtenção do total de 51 fragmentos polimórficos. A percentagem média de locos polimórficos, dentro de cada população, foi de 83,8%. A heterozigosidade corrigida de Nei esperada variou de 0,23 a 0,33, com média de 0,29, e o índice de diversidade genética de Shannon e Weaver variou de 0,29 a 0,48, com média de 0,42. No nível de espécie, estes dois índices apresentaram valores de 0,36 e 0,54, respectivamente. A análise de variância molecular mostrou que 66% da variância molecular total pode ser atribuída a diferenças intrapopulacionais e que os 34% restantes podem ser atribuídos a diferenças interpopulacionais. O teste de Mantel mostrou baixa correlação entre: distância geográfica e diferenciação genética, identidade genética e diferenciação genética e, distância genética de Nei e diferenciação genética. As populações brasileiras de Z. subfasciatus possuem baixa diferenciação genética e fraca estruturação geográfica.

The objective of this work was to evaluate the genetic diversity of populations of Zabrotes subfasciatus through ISSR molecular markers. Twelve populations from eight Brazilian states were sampled in the total of 269 individuals. Five ISSR primers were used and a total of 51 polymorphic bands were obtained. The percentage of polymorphism within population averaged 83.8%. Nei's unbiased expected heterozygosity ranged from 0.23 to 0.33, with an average of 0.29, and the Shannon & Weaver's index of gene diversity varied from 0.29 to 0.48, with an average of 0.42. At species level, these two indexes were 0.36 and 0.54, respectively. The analysis of molecular variance showed that 66% of the total molecular variance can be attributed to differences within population, while 34% was apportioned among populations. Mantel's test showed a low correlation between: geographic distance and genetic differentiation, genetic identity and genetic differentiation, and Nei's genetic distance and genetic differentiation. The genetic differentiation in Brazilian populations of Z. subfasciatus is low and geographic structuring is weak.

Psychotria ipecacuanha é uma espécie medicinal que se desenvolve em agregados perenes, denominados reboleiras, em áreas úmidas e sombrias no sub-bosque da Floresta Atlântica. O presente trabalho caracterizou a variação de atributos florais em 35 reboleiras de três populações naturais desta espécie. Observações de campo mostraram que as reboleiras são isomórficas, isto é, apresentam apenas uma forma floral (brevistila ou longistila). Estigmas e anteras estão posicionados reciprocamente em cada forma floral, um dimorfismo característico de distilia. As populações são isopléticas, isto é, apresentam razão equilibrada (1:1) entre as formas florais. Análises comparativas da morfometria floral revelaram que, independente da população investigada, as flores brevistilas apresentaram maiores médias de comprimento da antera, comprimento do estigma, diâmetro da corola e diâmetro do grão de pólen. Flores brevistilas apresentaram diferenças interpopulacionais significativas com relação aos atributos florais investigados. Flores da forma longistila também apresentaram diferenças interpopulacionais significativas, exceto quanto à altura dos estigmas e ao comprimento da corola. Polinizações controladas conduzidas em condições naturais mostraram que a produção de frutos foi maior após polinização legítima. Porém, observações de crescimento de tubos polínicos juntamente com a produção de frutos após autopolinização espontânea e polinização ilegítima sugerem que a espécie apresenta compatibilidade parcial intraforma.

Psychotria ipecacuanha is a perennial, medicinal herb that grows in clusters in the understory of humid, shady areas of the Atlantic Rain Forest of southeastern Brazil. The present study characterized the variation in floral traits among 35 clusters from three natural populations of this plant species. Field observations showed that the clusters are isomorphic, that is, a given cluster will either set long-styled or short-styled flowers. Stigmas and anthers are reciprocally placed in each morph, a dimorphism characteristic of distyly. The populations are isoplethic, that is, a given population exhibits an equilibrium 1:1 ratio of floral morphs. Morphometric analyses revealed that anther length, stigma length, corolla diameter, and pollen grain diameter were consistently greater in short-styled flowers, regardless of the population investigated. Significant differences for floral traits in the short-styled morph were found among populations. Floral traits in the long-styled morph also showed some significant differences among populations, but not for stigma height and corolla length. Controlled pollinations carried out in natural populations showed that fruit production was higher after inter-morph pollination. Nevertheless, observations of pollen tube growth in style, and also fruit production after spontaneous self-pollination and intra-morph pollination, indicated partial intramorph compatibility in this plant species.

This study aimed to analyse the content and the composition of the essential oil of five accesses of mentrasto (Ageratum conyzoides). Five accesses of mentrasto with three repetitions were used. The accesses were obtained in Mariana-MG (AMA), Piranga-MG (API), Visconde do Rio Branco (ARB) and Viçosa-MG (AVB and AVP). The essential oil was obtained by hydrodistillation and the identification of the oil components by CG and GC/MS. The ARB access presented the higher essential oil content, that is 0.70% (P < 0.05). Eleven chemical compounds were identified. The precocene I was the main constituent in the access API, and the precocene II was the main constituent in the accesses AMA, ARB, AVB and AVP.
